An OpenSTEM laboratory school providing you with 25 hours of ‘hands on’ experience of several widely used molecular techniques for quantifying gene expression. Western blot analysis is performed to examine protein levels with colorimetric and chemiluminescent visualization of HRP-linked secondary or biotin-streptavidin reagents. For RNA quantitation, you use qPCR approaches to quantitate mRNA in human cells: PCR efficiency, absolute quantitation against GAPDH calibration standards, cDNA synthesis by reverse transcription, relative quantitation against other commonly used reference genes using ΔCt analysis and dual label hydrolysis probe quantitation.
